Product Specific Information

MA5-16802 is suitable for use a negative control for measurement of non-specific binding of mouse monoclonals of isotype IgG1 in rat tissue in a dual labelling technique using FITC and R-Phycoerythrin fluorochromes. This antibody contains a monoclonal mixture of FITC-conjugated Mouse IgG1 RPE-conjugated Mouse IgG1.

Reconstitute with 0.5 mL of distilled water. After reconstitution, store product undiluted at 4°C in the dark.

For FACS analysis, use 10 µL of the suggested working dilution to label 1x10^6 cells in 100 µL.

Mouse anti Human CD8:FITC/CD4:RPE Dual Color reeagent may be used to simultaneously to identify CD4 and CD8 positive cells.

CD4, clone LT8 conjugated to FITC, is purified by ion exchange chromatography, and CD8, clone RPA-T4 conjugated to RPE, is prepared by affinity chromatography on Protein G and then purified by gel-filtration to separate the unconjugated material.

Target Information

CD4, a single chain transmembrane glycoprotein, is found on a T cell subset (helper/inducer) representing 45% of peripheral blood lymphocytes. It is also present on 80% of thymocytes and at a lower level on monocytes. It is involved in recognition of antigen presented along with MHC class II by APCs. It serves as receptor for HIV. The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen acts as a corepressor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The coreceptor functions as either a homodimer composed of two alpha chains, or as a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains.
