3 µg of total protein from Arabidopsis thaliana extracted with were separated on 15% SDS-PAGE and blotted 1 h to PVDF. Blots were blocked with 10% skimmed milk for 1 h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1:1,000 overnight at 4°C with agitation. Target Information
CGL40 encodes a small, basic nucleus-encoded chloroplast protein that is imported via an N-terminal transit peptide and accumulates in the thylakoid membrane as a low-molecular-weight component of photosystem I. The mature polypeptide is approximately 129 amino acids (about 14.7 kDa) and contains the characteristic photosystem I reaction center subunit N (PsaN) domain, consistent with a compact, lumen-exposed thylakoid protein rather than a large multi-pass membrane subunit. By contributing to photosystem I organization and/or stability at the thylakoid membrane, CGL40 supports efficient light-driven electron transfer in chloroplasts and thereby participates in photosynthesis, with expression detected across multiple plant tissues and developmental stages.
