          CNOT7 Antibody (PA5-101270) in ICC/IF

          Immunocytochemistry analysis of CNOT7 in Hela cells. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were incubated with CNOT7 polyclonal antibody (Product # PA5-101270) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies.           Antibody detects endogenous levels of total CNOT7.

          Target Information

          CNOT7 (CCR4-NOT transcription complex subunit 7) has 3'-5' poly(A) exoribonuclease activity for synthetic poly(A) RNA substrate. Its function seems to be partially redundant with that of CNOT8. CNOT7 is a catalytic component of the CCR4-NOT complex, which is one of the major cellular mRNA deadenylases, and is linked to various cellular processes including bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation, and general transcription regulation. During miRNA-mediated repression, the complex seems to act as a transcriptional repressor during translational initiation, as well. Additional complex functions may be a consequence of its influence on mRNA expression. CNOT7 associates with members of the BTG family such as TOB1 and BTG2 and is required for their anti-proliferative activity.
