Immunofluorescence analysis of Caspase 3 was done on 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Caspase 3 [3CSP03 (4.1.18)] Rabbit Polyclonal Antibody (PA516335) at 2ug/ml in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d is a merged image showing nuclear localization. Panel e is a no primary antibody control. The images were captured at 60X magnification.

PA5-16335 targets Caspase 3 in immunohistochemistry (paraffin), immunoprecipitation, and western blot applications and shows reactivity with Bovine, Canine, Hamster, Human, mouse, Non-human primate, Ovine, Porcine, Rabbit, and Rat samples.
The PA5-16335 immunogen is recombinant full length human caspase 3 protein.
Caspase 3 is ubiquitously expressed and like other caspases is synthesized as an inactive (32kDa) proenzyme. Upon activation, Caspase 3 is cleaved at Asp28-Ser29 and Asp175-Ser176 thereby generating two subunits of 17kDa and 12kDa, respectively. Recent studies have implicated that Caspase 3 is associated with the induction of apoptosis. Activation of Caspase 3 occurs in response to variety of apoptotic inducers including Fas mediated apoptosis.
