Target Information
CpxA is a bacterial inner-membrane, envelope-stress sensor histidine kinase that spans the cytoplasmic membrane with two transmembrane helices flanking an extracytoplasmic/periplasmic sensor region and a cytosolic transmitter module; the periplasmic sensor adopts a PAS-like (Per-ARNT-Sim) fold and dimerizes as part of signaling control, while the cytoplasmic portion contains the canonical histidine-kinase core (including HAMP, dimerization/phosphotransfer and catalytic/ATP-binding subdomains) that supports autophosphorylation and phosphotransfer. Functionally, CpxA forms a two-component system with the response regulator CpxR, relaying envelope perturbations such as misfolded extracytoplasmic proteins by modulating CpxR phosphorylation state to reprogram transcription of cell-envelope quality-control and biogenesis factors; in Escherichia coli, Cpx pathway activation increases expression of periplasmic folding and degradation machinery including DsbA (disulfide bond formation), DegP (periplasmic protease/chaperone), and peptidyl-prolyl isomerases, thereby maintaining envelope proteostasis and promoting adaptation to extracytoplasmic stress.
