A member of DAP kinase family, DAPK2 has an N-terminal protein kinase domain, a conserved CAM binding domain and a 40A.A C-terminal tail required for homodimerization. Inactive DAPK2 is phosphorylated at conserved and quote;Ser-308 and quote;, an inhibitory autophosphorylation site located within the CAM binding domain and is critical in inhibiting DAPK2 catalytic activation. Pro-apoptotic function of DAPK2 is activated by dephosphorylation, increase of intracellular Ca2+ levels, Ca2+ induced Calmodulin binding and homodimerization. Reports suggest DAPK2 along with DAPK1 induces membrane blebbing and formation of autophagic vesicles and plays a vital role in TGF-induced caspase-dependent Type-I and IFNã- induced caspase-independent autophagic type II cell death signaling.
