DRAM is an important lysosomal membrane protein of the p53 tumor suppressor pathway. It is required for the induction of macroautophagy and as an effector of p53-mediated death. DRAM expression is induced due to cellular stress via endogenous p53 leading to bulk degradation of cytoplasmic components. It is necessary but not sufficient for apoptotic cell death from p53, while over expression of DRAM alone causes minimal cell death. Quantitative PCR revealed that DRAM was significantly down regulated in tumor cell lines by methylation-dependent transcriptional silencing, particularly in cancers lacking other mechanisms of p53 inactivation.
