DVL3 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of DVL3 was performed using 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with anti-DVL3 Rabbit Polyclonal Antibody (Product # PA5-17482) at 1:100 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoplasmic localization. Panel e represents cells treated with 1mM Rapamycin for 24 hours showing downregulation of DVL3 expression, while panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

DVL3 Antibody in Western Blot (WB) — Western blot analysis was performed on whole cell extracts of A-375 (Lane 1), K-562 (Lane 2), HCT 116 (Lane 3), MCF-7 (Lane 4), MCF-7 treated with AICAR (2mM for 24 hr) (Lane 5), MCF-7 treated with Metformin (40mM for 24hr) (Lane 6), HT-29 (Lane 7), HeLa treated with Rapamycin (1nM for 24hr) (Lane 8), HeLa (Lane 9) and Mouse Brain (Lane 10). The blot was probed with DVL3 Polyclonal Antibody (Product # PA5-17482, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 90kDa band corresponding to DVL3 was observed across the panel and was downregulated upon AICAR and Metformin treatment in MCF-7 cells and Rapamycin treatment in HeLa cells.

DVL3 Antibody — Altered expression of proteins upon cell treatment demonstrates antibody specificity. Western blot using DVL3 polyclonal antibody (Product # PA5-17482), shows decreased expression of proteins in MCF-7 upon AICAR and Metformin treatment and decreased expression in HeLa upon Rapamycin treatment. {TM}
