Western blot analysis of FOXA3 was performed by loading 25 ug of HepG2 (lane 1) mouse liver (lane 2) and rat liver (lane 3) lysates onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4°C overnight. The membrane was probed with a FOXA3 polyclonal antibody (Product # PA1-813) at a dilution of 1:500 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Plus Western Blotting Substrate (Product # 32132). Results show a band at ~37kDa using an exposure time of 2 min (HepG2, mouse liver) and 30 sec (rat liver).

PA1-813 has been successfully used in Western blot and ChIP applications. By Western blot, PA1-813 detects a ~37 kDa band representing FOXA3 from mouse liver.
