Western blot analysis of FTO in extracts of various cell lines. Samples were incubated with FTO Polyclonal antibody (Product # PA5-120656) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 180s.

Target Information
FTO is a novel protein and a member of the non-heme dioxygenase (Fe(II)-and 2-oxoglutarate-dependent dioxygenases) superfamily. Amino acid conservation patterns support this hypothesis and indicates that both 2-oxoglutarate and iron should be important for FTO function. Though not much is yet known about FTO, it is thought that the protein may somehow modify the activity of genes involved in metabolism and fat storage, which in turn may influence a person's risk of obesity.
