GLUT4 Antibody (PA5-23052) in WB

Western blot was performed using Anti-GLUT4 Polyclonal Antibody (Product # PA5-23052) and due to heavy glycosylation, bands between 40-260 kDa corresponding to GLUT4 were observed across tissues tested. Also, the band was observed in 3T3-L1 differentiated to adipocytes when compared to undifferentiated 3T3-L1. The expression of GLUT4 is reported to be high in adipocytes. GLUT4 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of GLUT4 was performed using 70% confluent log phase 3T3-L1 cells differentiated to adipocytes. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with GLUT4 Rabbit Polyclonal Antibody (Product # PA5-23052) at 5 µg/mL in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing cytoplasmic & membrane localization. Panel e represents 3T3-L1 (undifferentiated) cells having no expression of GLUT4. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

GLUT4 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of GLUT4 in mouse kidney. Samples were incubated in GLUT4 polyclonal antibody (Product # PA5-23052).

GLUT4 Antibody in Flow Cytometry (Flow) — Flow cytometry of GLUT4 in HepG2 and a matched isotype control. Samples were incubated in GLUT4 polyclonal antibody (Product # PA5-23052) using a dilution of 5 µg/mL for 30 minutes at room temperature followed by a Rabbit IgG (H+L) Cross-Adsorbed secondary antibody. Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin.

Target Information

This gene is a member of the solute carrier family 2 (facilitated glucose transporter) family and encodes a protein that functions as an insulin-regulated facilitative glucose transporter. In the absence of insulin, this integral membrane protein is sequestered within the cells of muscle and adipose tissue. Within minutes of insulin stimulation, the protein moves to the cell surface and begins to transport glucose across the cell membrane. Mutations in this gene have been associated with noninsulin-dependent diabetes mellitus (NIDDM).
