Seedlings were frozen in liq N 2, then lysed with tissue lyzer, and 100 µL extraction buffer (100 m M Na H 2 PO 4 10 m M Tris/HCl p H 8.0,8 M Harnstoff) was added. Target Information
GNOM encodes a large, membrane-associated ARF guanine nucleotide exchange factor that cycles between cytosol and endomembranes within the plant endomembrane system, where it has been detected on endosomal compartments and, in imaging-based analyses, prominently at the Golgi apparatus. The protein is a Sec7-domain ARF-GEF (brefeldin A sensitive), with a conserved catalytic Sec7 domain that activates ARF-family small GTPases to drive vesicle budding and trafficking; genetic and biochemical data also support GNOM self-association via an N-terminal conserved region important for subunit interaction. Functionally, GNOM-dependent trafficking is essential for establishing and maintaining cell polarity during embryogenesis and for auxin-dependent development: loss of GNOM disrupts apical-basal patterning and causes auxin-related growth defects, while inhibition of GNOM activity perturbs the polar localization and recycling of PIN auxin efflux carriers (notably PIN1), thereby impairing directional auxin transport and downstream organogenesis.
