Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human HB-EGF. Anti-Human HB-EGF-specific antibody was purified by affinity chromatography employing an immobilized Human HB-EGF matrix.
Sandwich ELISA: To detect Human HB-EGF by sandwich ELISA (using 100 µL/well), a concentration of 0.25-1.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Rabbit Anti-Human HB-EGF (500-P329BT) as a detection antibody, allows the detection of at least 2000-4000 pg/mL of Recombinant Human HB-EGF.
Western Blot: To detect Human HB-EGF by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. When used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human HB-EGF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
HBEGF is a growth factor that mediates its effects via EGFR, ERBB2 and ERBB4. It is required for normal cardiac valve formation and normal heart function. HBEGF promotes smooth muscle cell proliferation. It may be involved in macrophage-mediated cellular proliferation. It is mitogenic for fibroblasts, but not endothelial cells. It is able to bind EGF receptor/EGFR with higher affinity than EGF itself and is a far more potent mitogen for smooth muscle cells than EGF. It also acts as a diphtheria toxin receptor.
