Immunofluorescent staining of Rat PC12 cells using PA5-19494, anti-HIF2 alpha antibody. The cells were fixed with PFA (4%)for 10 minutes, permabilised PBS-T for 20 minutes and exposed to the primary antibody at a concentration of 1 ug/ml overnight at 4C. A solution of BSA (1%), normal goat serum (10%) and glycine (0.3 M) was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody was a 448 fluorescence conjugated Goat anti-rabbit IgG (green) at a dilution of 1:1000. A WGA- 594 fluorescent conjugated stain was used to label plasma membranes (red) and the nuclei stain was DAPI (blue).

For Western Blot, this antibody detects a HIF-2-alpha specific band at ~96 kDa and has non-specific bands at 240 kDa,25 kDa and 45 kDa.

This antibody is predicted to react with mouse based on sequence homology.

This gene encodes a transcription factor involved in the induction of genes regulated by oxygen, which is induced as oxygen levels fall. The encoded protein contains a basic-helix-loop-helix domain protein dimerization domain as well as a domain found in proteins in signal transduction pathways which respond to oxygen levels. Mutations in this gene are associated with erythrocytosis familial type 4.
