HMGCR Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of HMGCR was performed using 70% confluent log phase T-47D cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with HMGCR Polyclonal Antibody (Product # PA5-37367) at 1:200 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790), (1:2000), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing ER and peroxisomal membrane localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

HMGCR Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of HMGCR in paraffin-embedded mouse liver cancer tissue. Samples were incubated with HMGCR polyclonal antibody (Product # PA5-37367).

HMGCR Antibody in Western Blot (WB) — Western Blot was performed using Anti-HMGCR Polyclonal Antibody (Product # PA5-37367) and a 100 kDa band corresponding to Pro-HMGCR and a 55 kDa cleaved isoform were observed across cell lines tested except MDA-MB-231. Membrane enriched extracts (30 µg lysate) of MCF7 (Lane 1), MCF7 treated with 10 µM Atorvastatin for 48 hours (Lane 2), T-47D (Lane 3), T-47D treated with 10 µM Atorvastatin for 48 hours (Lane 4), MDA-MB-231 (Lane 5) and MDA-MB-231 treated with 10 µM Atorvastatin for 48 hours (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Uncharacterized bands (*) were observed around ~35, 70, 82 kDa.

Target Information

Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. Grik2, also known as glutamate receptor 6, belongs to the kainate family of glutamate receptors, which are composed of four subunits and function as ligand-activated ion channels. Recent reports have suggested that defects in the Grik2 protein may be associated with autosomal recessive mental retardation and possibly other neurological disorders such as schizophrenia. Numerous isoforms of Grik2 are known to exist and may be subject to RNA editing within the second transmembrane domain, which is thought to alter the properties of ion flow. This Grik2 antibody may exhibit some cross-reactivity to Grik3.
