Western blot analysis of IFI16 was performed by loading 30 µg of THP-1 lysate in reducing sample buffer onto a 4-15% Precast Protein Gels. Proteins were transferred to PVDF membrane and followed by blocking in TBST+5% non-fat milk for one hour at room temperature. IFI16 was detected at approximately 88 kDa using an IFI16 polyclonal antibody (Product # PA5-39180) at a dilution of 1:500 overnight at 4°C on a rocking platform, followed by a Rabbit IgG Horseradish Peroxidase-conjugated Antibody at a dilution of 1:1,000 for one hour. Chemiluminescent detection was performed using SuperSignal™ West Pico Chemiluminescent Substrate (Product # 34080) and ImageQuant LAS 4000 (GE, 28-9558-10).

This gene encodes a member of the HIN-200 (hematopoietic interferon-inducible nuclear antigens with 200 amino acid repeats) family of cytokines. The encoded protein contains domains involved in DNA binding, transcriptional regulation, and protein-protein interactions. The protein localizes to the nucleoplasm and nucleoli, and interacts with p53 and retinoblastoma-1. It modulates p53 function, and inhibits cell growth in the Ras/Raf signaling pathway.
