LMX1A Antibody in Western Blot (WB) — Western blot analysis was performed on tissue extracts (30 µg lysate) of Rat Brain (Lane 1), Mouse Brain (Lane 2), Mouse Cerebellum (Lane 3) and Mouse heart (Lane 4). The blots were probed with Anti-Lmx1a Recombinant Rabbit Polyclonal Antibody (Product # 710980, 1-2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). A 42 kDa band corresponding to Lmx1a was observed according to the tested tissues. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12% Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western blotting Substrate (Product # 32106).

LMX1A Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell models owing to their inherent genetic constitution. Expression of LMX1A was observed specifically in rat brain, mouse brain and mouse cerebellum, but not in mouse heart, using Anti-LMX1A Recombinant Rabbit Polyclonal Antibody (Product # 710980) in Western blot. LMX1A is a transcriptional activator, which is expressed in the central nervous system, but is absent in the heart. {RE}

Target Information

The LIM homeobox transcription factor 1 (LMX1A) belongs to the LIM-homeodomain family. Members of this family are known to be important for pattern formation during development. LMX1A binds an A/T-rich sequence and functions in the nucleus as a transcriptional activator to the insulin gene promoter. In the developing embryo, LMX1A is expressed along the neuraxis and leads to the development of the roof plate of the vertebrae. While LMX1A is crucial for the differentiation of dopamine neurons, the related LMX1B appears to be important for both the development and the survival of these cells. It has been suggested that LMX1A might be a useful biomarker or risk factor in tumor aggressiveness.
