VDAC Antibody (MA5-45668) in ICC/IF
Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% PFA for 15 min. Samples were incubated with VDAC1 monoclonal antibody (Product # MA5-45668) at 1:100 for overnight at 4°C with slow rocking, followed by AlexaFluor 488 at 1:1,000 for 1 hour at RT. Counterstain used was Phalloidin-iFluor 647 (red) F-Actin stain; Hoechst (blue) nuclear stain at 1:800, 1.6mM for 20 min at RT. (A) Hoechst (blue) nuclear stain. Mouse: 98% identity (279/283 amino acids identical). Rat: 98% identity (279/283 amino acids identical) >60% identity with VDAC2 and VDAC3.
1 µg/mL of MA5-45668 was sufficient for detection of VDAC1 in 20 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.|Detects approximately 30kDa. Does not cross-react with VDAC2 or VDAC3 (based on KO validation results).

Target Information
Voltage-dependent anion channel (VDAC) proteins are abundant, pore-forming proteins belonging to the eukaryotic mitochondrial porins. It was discovered in the mitochondrial outer membrane. It also is expressed in the plasma membrane. At least three different VDAC genes have been identified in vertebrates. VDAC proteins are known to play an essential role in cellular metabolism and in the early stages of apoptosis. For example, VDAC constitutes a major pathway by which metabolites such as ADP/ATP, succinate and citrate are exchanged between the cytosol and mitochondria. And VDAC1 in the plasma membrane establishes a novel level of apoptosis regulation putatively via its redox activity.
