Immunocytochemistry/Immunofluorescence analysis using human neuroblastoma cells. Fixation involved 4% Formaldehyde for 15 min at RT. Samples were incubated with GABA-A Receptor Alpha4 monoclonal antibody (Product # MA5-45736) at 1:100 for 60 min at RT, followed by Goat Anti-Mouse ATTO 488 at 1:100 for 60 min at RT. A 1:100 dilution of MA5-45736 was sufficient for detection of GABA-A R, Alpha4 in 20 µg of mouse brain lysate by ECL immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody. |Detects approximately 60kDa. Does not cross-react with other GABA-A-Receptor Alpha subunits.
This antibody was formerly sold as clone S398A-34.
Target Information
Gamma-aminobutyric acid is the major inhibitory neurotransmitter in the mammalian brain where it acts at GABA-A receptors, which are ligand-gated chloride channels. Chloride conductance of these channels can be modulated by agents such as benzodiazepines that bind to the GABA-A receptor. At least 16 distinct subunits of GABA-A receptors have been identified. This gene encodes subunit alpha-4, which is involved in the etiology of autism and eventually increases autism risk through interaction with another subunit, gamma-aminobutyric acid receptor beta-1. Alternatively spliced transcript variants encoding different isoforms have been found in this gene.
