Modafinil is a wake-promoting agent primarily targeting the dopamine transporter (DAT), leading to reuptake inhibition and increased dopamine levels in the synaptic cleft. Structurally, modafinil is characterized by a sulfoxide functional group, which has been modified in various analogues to enhance biological activity. These analogues have shown potential in cognitive enhancement and treatment of psychostimulant use disorders. Modafinil derivatives have been synthesized through substitution and coupling reactions, demonstrating anti-inflammatory effects by inhibiting nitrite production and expression of iNOS and COX-2 in cultured BV2 cells.
