Immunofluorescent analysis of NOX4 (green) HEK293T cells. Cells fixed with 4% formaldehyde were permeabilized and blocked with 1X PBS containing 5% BSA and 0.3% Triton X-100 for 1 hour at room temperature. Cells were probed with a NOX4 polyclonal antibody (Product # PA1-46014) at a dilution of 1:100 overnight at 4°C in 1X PBS containing 1% BSA and 0.3% Triton X-100, washed with 1X PBS, and incubated with a fluorophore-conjugated goat anti-rabbit IgG secondary antibody at a dilution of 1:200 for 1 hour at room temperature. In Western blot, this antibody recognizes a band at ~67 kDa or larger representing isoform 1 of NOX4, and what appears to be a non-specific band ~48 kDa.
The observed band size may vary depending on sample type and glycosylation.
Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.Suggested positive control: whole normal kidney tissue lysate.
Oxygen sensing is essential for homeostasis in all aerobic organisms. A phagocyte-type oxidase, similar to that responsible for the production of large amounts of reactive oxygen species (ROS) in neutrophil granulocytes, with resultant antimicrobial activity, has been postulated to function in the kidney as an oxygen sensor that regulates the synthesis of erythropoietin in the renal cortex. NOX4 has a role as a redox messenger in the activation of intracellular signaling pathways leading (or contributing) to mitochondriogenesis, cell survival, and differentiation in hematopoietic stem cells. Data suggest that NOX4 provides a novel link between the insulin receptor and the generation of cellular reactive oxygen species that enhances insulin signal transduction.
