PA1-755 detects prostate apoptosis response protein (Par-4) from human and rat tissues.
PA1-755 has been sucessfully used in Western blot procedures. By Western blot, this antibody detects an ~40 kDa protein representing Par-4 in AT-3 cell extract. In Western blots of rat tissue extracts, including testes, spleen, ovary, lung, and skeletal muscle, ~40 kDa and ~38 kDa proteins representing Par-4 are detected. In Western blots of T47D cell extracts, an ~40 kDa protein representing Par-4 and a smaller ~20 kDa unknown protein is detected.
PA1-755 immunizing peptide corresponds to amino acids residues 324-342 from human Par-4. PA1-755 immunizing peptide (Cat. # PEP-060) is available for use in neutralization and control experiments.
This gene encodes a tumor suppressor protein that selectively induces apoptosis in cancer cells through intracellular and extracellular mechanisms. The intracellular mechanism involves the inhibition of pro-survival pathways and the activation of Fas-mediated apoptosis, while the extracellular mechanism involves the binding of a secreted form of this protein to glucose regulated protein 78 (GRP78) on the cell surface, which leads to activation of the extrinsic apoptotic pathway. This gene is located on the unstable human chromosomal 12q21 region and is often deleted or mutated different tumors. The encoded protein also plays an important role in the progression of age-related diseases.
