Detection of human PBRM1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PBRM1 antibody (Product # A301-590A) (lot 2) and rabbit anti-PBRM1 antibody A301-591A. For blotting immunoprecipitated PBRM1, Rabbit anti-PBRM1 recombinant monoclonal antibody A700-019 was used at 1:1,000. Detection: Chemiluminescence with an exposure time of 30 seconds.

Target Information
The PBRM1 gene encodes a subunit of the PBAF (Polybromo-associated BAF) chromatin remodeling complex, playing a crucial role in the regulation of chromatin structure and transcription. PBRM1, also known as BAF180, contains bromodomains that recognize acetylated lysine residues on histone tails, facilitating chromatin remodeling necessary for effective transcriptional regulation. This gene is frequently mutated in renal cell carcinoma, particularly clear cell renal cell carcinoma (ccRCC), where such mutations are implicated in the pathogenesis of the disease. PBRM1 acts as a tumor suppressor, and its loss of function can lead to deregulation of gene expression, contributing to oncogenic processes. It is involved in pathways that govern cell cycle regulation and DNA damage response, reflecting its importance in maintaining genomic stability.
