PDE4D3 (Phosphodiesterase 4D, cAMP-specific, isoform 3) is an enzyme encoded by the PDE4D gene, which is located on chromosome 5. PDE4D3 belongs to the PDE4 family, which plays a crucial role in hydrolyzing cyclic AMP (cAMP), thereby regulating intracellular levels of this important signaling molecule. PDE4D3 is integral in modulating cAMP signaling pathways, impacting processes such as cardiac function and inflammation. This isoform of phosphodiesterase has been found to interact within a large protein complex, including Epac1, PKA, and ERK5, which is anchored by mAKAP. Phosphorylation by ERK decreases PDE4D3 activity, leading to local cAMP accumulation and increased activation of cAMP-dependent pathways.
