PIAS1, PIAS2, and PIAS3 are members of the Protein Inhibitor of Activated STAT (PIAS) family, which are integral to cellular processes such as SUMOylation, transcriptional regulation, and signal transduction. PIAS1 is noted for its role as a chromatin-bound androgen receptor (AR) coregulator, influencing AR-mediated target gene regulation in prostate cancer cells. It interacts with and modifies AR via SUMO3, which affects cell proliferation. PIAS2, another member, has been implicated in the inhibition of the JAK-STAT signaling pathway, notably suppressing STAT4-dependent gene induction linked to IL-12. Furthermore, PIAS2 has been shown to repress IFN-B production, highlighting its regulatory role in immune response mechanisms. PIAS3 is recognized as a SUMO E3 ligase that binds activated STAT3 and inhibits its DNA binding ability, subsequently hindering STAT3-mediated gene activation. Elevated expression of PIAS3 in prostate cancer samples has been associated with apoptosis induction. Together, the PIAS family genes are increasingly acknowledged in cancer biology due to their contributions to growth regulation and metastasis, while also representing potential therapeutic targets in cancer treatment strategies.
