Immunofluorescence analysis of PMS2 was performed using 70% confluent log phase HeLa and DU 145 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. PMS1 protein homologue 2 (PMS2) is a DNA mismatch repair protein. Defects in PMS2 are a cause of hereditary non-polyposis cancer (HNPCC). Human PMS2 is an amino acid protein of 862 amino acids.
