75 µg of total protein from Solanum tuberosum v. Nicola extracted with 20 m M TRIS PH-8.5,10 m M thiourea, 10 m M Ca Cl 2,5 m M DTT, 1 m M PMSF, 1%PVPP were separated on 4-20% SDS-PAGE and blotted 1.5 h to PVDF. Blots were blocked with 5% milk powder in T-TBS (2.5 gr of powder in 50 mL T-TBS) for 1 h at room temperature (RT) with agitation. As a storage protein, patatin provides nitrogen and carbon during sprouting and early growth, while its enzymatic activity can modulate membrane integrity and metabolic pathways. Variants show tissue specific expression and differing catalytic properties, and manipulation of patatin levels affects tuber quality traits, defense capacity, and responses to environmental challenges.
