Prolactin Receptor Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of Prolactin Receptor on paraffin-embedded human mammary cancer tissue. Sample was incubated with Prolactin Receptor polyclonal antibody (Product# PA5-79876).

Prolactin Receptor Antibody in Western Blot (WB) — Western blot analysis of PRLR in, Lane 1: human Hela whole cell lysates, Lane 2: human MCF-7 whole cell lysates. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50 µg of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. The membrane was blocked with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with Prolactin Receptor Polyclonal Antibody (Product # PA5-79876) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0. 1% Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit. A specific band was detected for PRLR at approximately 95KD. The expected band size for PRLR is at 70KD.

Target Information

Prolactin (PRL) is a pituitary hormone involved in the stimulation of milk production, salt and water regulation, growth, development and reproduction. PRL exerts its influence on different cell types through a signal transduction pathway which begins with the binding of the hormone to a transmembrane PRL receptor. Immunoreactive PRL receptor (PRLR), a member of the cytokine receptor family, varies in size (short and long forms) with tissue source and species, from ~40 kDa to 100 kDa. PRLR consists of at least three separate domains: an extracellular region with 5 cysteines which contains the prolactin binding site, a single transmembrane domain and a cytoplasmic region, the length of which appears to influence ligand binding and regulate cellular function.
