RPE65 Antibody (PA5-115852) in ICC/IF
Immunocytochemistry analysis of RPE65 in HeLa cells. Samples were treated with PFA, permeabilized in 0.1% Triton X-100, blocked in 10% serum (45 min at 25°C), and incubated with polyclonal antibody (Product # PA5-115852) at a dilution of 1:200 (1 hr at 37°C). Secondary staining was applied with mouse anti-beta tubulin; AlexaFluor 594 conjugated goat anti-rabbit IgG (Red); AlexaFluor 88 conjugated goat anti-mouse IgG (Green) and DAPI (blue) using a dilution of 1:200 (1 hr at 37°C).

Antibody detects endogenous levels of total RPE65.
Target Information
RPE65 is a major protein of the retinal pigment epithelium (RPE). RPE65 is essential for the regeneration of rhodopsin in the visual cycle. Mutations in RPE65 are responsible for certain forms of autosomal recessive severe retinal dystrophy, including Leber Congenital Amaurosis (LCA). Loss of function results in a block in retinoid processing required for the synthesis of 11-cis retinal and the accumulation of retinyl esters in the RPE.
