PA1-815 detects retinoid X receptor (RXR) beta from human and mouse cells as well as transfected human and mouse RXR beta.
PA1-815 has been successfully used in Western blot, immunoprecipitation and gel shift procedures. By Western blot, this antibody detects an ~50 kDa protein representing RXR beta from COS cells transfected with the human gene. Gel shift assays reveal supershifting of vitamin D receptor (VDR)/RXR beta/vitamin D receptor response element (VDRE) complexes when PA1-815 is preincubated with RXR beta prior to addition of VDR.
The PA1-815 immunogen is a synthetic peptide corresponding to residues Q(190) K S D Q G V E G P G A T(202) of mouse RXR beta.
Recombinant human VDR (Cat. # RP-710) and human RXR beta (catalog # RP-812) proteins can be purchased separately for control experiments in Western blot, immunoprecipitation and gel shift procedures.
Retinoid X receptors (RXRs) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptor proteins which exert their effects by binding to specific DNA response elements, thus regulating gene expression in target cells. The RXR subfamily consists of at least three similar genes, RXR alpha, RXR beta and RXR gamma all of which control transcription of target genes mediated by retinoids. RXR beta controls expression of many genes that respond to hormones and vitamins, including thyroid hormone, estrogen, retinoids and vitamin D. RXR beta, also known as RCoR-1, controls such a wide array of genes because of its ability to heterodimerize with other hormone receptors including the thyroid hormone receptor (THR), vitamin D receptor (VDR) and retinoic acid receptor (RAR).
The corresponding gene for the retinoid X receptor beta is NR2B2.
