S100A10 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of S100A10 in A431 cell. Antigen retrieval was performed on the tissue using IHC enzyme antigen retrieval reagent and blocked with 10% goat serum. Samples were incubated with S100A10 polyclonal antibody (Product # PA5-95505) at a 2 µg/1x10^6 cells dilution, followed by 488 conjugated goat anti-rabbit IgG using a 1:100 dilution and DAPI.

S100A10 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of S100A10 in paraffin-embedded human mammary cancer tissue. Antigen retrieval was performed on the tissue using citrate buffer (pH 6, 20 min) and blocked with 10% goat serum. Samples were incubated with S100A10 polyclonal antibody (Product # PA5-95505) at a 1 µg/mL dilution, followed by biotinylated goat anti-rabbit IgG (30 min, 37°C), and developed with Strepavidin-Biotin-Complex and DAB.

S100A10 Antibody in Western Blot (WB) — Western blot was performed using Anti-S100A10 Polyclonal Antibody (Product # PA5-95505) and a 11 kDa band corresponding to Protein S100-A10 was observed across the panel tested except SW480 which is reported to be negative. Whole cell extracts (30 µg lysate) of HaCaT (Lane 1), HeLa (Lane 2), PANC-1 (Lane 3), HT-29 (Lane 4), SW480 (Lane 5), NIH:OVCAR-3 (Lane 6), NIH:OVCAR-3 (treated with Retinoic acid, 5 µm for 6days) (Lane 7) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

S100A10 Antibody in Flow Cytometry (Flow) — Flow cytometry of S100A10 in A549 cells (blue line), isotype control rabbit IgG (green line) and unlabeled (red line). Samples were blocked with 10% goat serum, incubated with S100A10 polyclonal antibody (Product # PA5-95505) at a dilution of 1 µg (per 1x10^6 cells), followed by 488 conjugated goat anti-rabbit IgG (30 min at 20°C) using a 5-10 µg (per 1x10^6 cells) dilution.

S100A10 Antibody in Western Blot, Immunocytochemistry (WB, ICC/IF) — Figure 6 Melatonin reversed the increased expression of C3, and decreased expression of S100A10 in primary astrocytes stimulated by IL-1alpha, TNF-alpha and C1q in vitro. After melatonin treatment and IL-1alpha, TNF-alpha and C1q exposure, the astrocytes were processed for immunofluorescence staining using primary antibodies GFAP (green), and anti-C3 (red) for A1 detection. Immunofluorescence images of cultured primary astrocytes showing the expression of GFAP and C3 at 24h after the treatment with IL-1alpha + C1q + TNF-alpha, IL-1alpha + C1q + TNF-alpha + melatonin and IL-1alpha + C1q + TNF-alpha + melatonin + luzindole when compared with the corresponding control ( A ). S100A10 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines tested owing to their inherent genetic constitution. Target Information

The S-100 family of calcium-activated proteins interact with a range of target proteins to modulate biological signaling pathways. Numerous cancer cell lines overexpress the plasminogen receptor S-100A10 on the extracellular cell surface, where it forms a heterotetrameric complex with Annexin II, though this association is not required for plasma membrane localization or binding and activation of plasminogen. Additionally, S-100A10 acts as a cellular chaperone for hepatitis B (Hep B) virus polymerase. Hep B virus polymerase normally localizes to the cytoplasm only, though in the presence of S-100A10 a portion relocates to the nucleus, implying a role for S-100A10 and intracellular calcium in the process of viral replication.
