S6K1/2 encode closely related AGC-family serine/threonine kinases that act predominantly in the cytoplasm but can also be detected in the nucleus in an isoform- and phosphorylation-dependent manner. They are soluble proteins (no transmembrane segments) built around a conserved central protein kinase catalytic domain flanked by divergent N- and C-terminal regulatory regions; activation requires multisite phosphorylation, including hydrophobic-motif phosphorylation (classically at Thr389 in S6K1) that promotes the active kinase conformation and facilitates activation-loop phosphorylation by PDK1. Functionally, S6K1 and S6K2 are major downstream effectors of mTORC1 signaling: once activated, they phosphorylate ribosomal protein S6 and additional translation-related substrates such as eIF4B, thereby promoting ribosome biogenesis and selective translation programs (including 5prime TOP mRNAs), which drives anabolic growth, increased protein synthesis, and cell proliferation; consistent with this role, altered S6K signaling is frequently linked to growth control and cancer-relevant phenotypes.
