Immunofluorescent analysis of SCAP in Hela cells. Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min) and probed with a SCAP polyclonal antibody (Product # PA5-35341) at a dilution of 1:100 for 2 hours at room temperature, followed by detection using a fluorescent conjugated secondary antibody (green) (1:1000, 1h). Nuclei were stained with Hoechst 33342 (blue) (10 ug/ml, 5 min).

SCAP is a protein with a sterol sensing domain(SSD) and seven WD domains. In the presence of cholesterol, this protein binds to sterol regulatory element binding proteins (SREBPs) and mediates their transport from the ER to the Golgi. The SREBPs are then proteolytically cleaved and regulate sterol biosynthesis.
