SIGLEC7 Antibody (PA5-47079) in Neu
Neutralization of SIGLEC7 in human red blood cells. Samples were incubated in SIGLEC7 polyclonal antibody (Product # PA5-47079). Recombinant Human Siglec‚7 Fc Chimera, immobilized onto a microplate, supports the adhesion of human red blood cells in a dose-dependent manner (orange line). Adhesion elicited by Recombinant Human Siglec‚7 Fc Chimera (1.5 µg/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human Siglec‚7 Antigen Affinity-purified Polyclonal Antibody. The ND50 is typically 0.3-0.9 µg/mL.

In direct ELISAs, approximately 15% cross-reactivity with recombinant human (rh) Siglec-9 is observed and approximately 5% cross-reactivity with recombinant mouse Siglec-E, rhSiglec-6, and rhSiglec-8 is observed.
Reconstitute at 0.2 mg/mL in sterile PBS.
Endoxin level is <0.10 EU per 1 µg of the antibody by the LAL method.
Target Information
Putative adhesion molecule that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,3- and alpha-2,6-linked sialic acid. Also binds disialogangliosides (disialogalactosyl globoside, disialyl lactotetraosylceramide and disialyl GalNAc lactotetraoslylceramide). The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Mediates inhibition of natural killer cells cytotoxicity. May play a role in hemopoiesis. Inhibits differentiation of CD34+ cell precursors towards myelomonocytic cell lineage and proliferation of leukemic myeloid cells (in vitro).
