Immunofluorescent analysis of SMAD4 showing staining in the cytoplasm and nucleus of Hela cells using a SMAD4 polyclonal antibody (Product # PA5-35330) followed by detection using a fluorescent conjugated secondary antibody (green). Cytoplasmic actin was stained with a fluorescent red phalloidin (7units/ml, 1 h at 37°C). Nuclei were stained with DAPI (blue) (10 ug/ml, 10 min).

SMAD4 is the common SMAD (co-SMAD)mediator of signal transduction by TGF-beta (transforming growth factor). It promotes binding of the SMAD2/SMAD4/FAST-1 complex to DNA and provides an activation function required for SMAD1 or SMAD2 to stimulate transcription. It may act as a tumor suppressor.
