SPT5 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescent staining of SPT5 in human cell line A-431 shows positivity in nucleus but excluded from the nucleoli. Samples were probed using a SPT5 Polyclonal Antibody (Product # PA5-56100).

SPT5 Antibody in Immunohistochemistry (IHC) — Immunohistochemical staining of SPT5 in human testis using a SPT5 Polyclonal Antibody (Product # PA5-56100) shows moderate nuclear positivity in cells in seminiferous ducts.

SPT5 Antibody in Western Blot (WB) — Western blot was performed using Anti-SPT5 Polyclonal Antibody (Product # PA5-56100) and a 150 kDa band corresponding to SPT5 was observed across all the cell lines tested. Modified whole cell extracts (1% SDS) (30 µg lysate) of HeLa (Lane 1), Hep G2 (Lane 2), NIH/3T3 (Lane 3) and K-562 (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by XCell SureLock™ Mini-Cell and XCell II™ Blot Module (Product # EI0002). The blot was probed with the primary antibody (0.4 ug/ml) and detected by chemiluminescence with Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

SPT5 Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. HeLa cells were transfected with SPT5 siRNA and decrease in signal intensity was observed in western blot application using Anti-SPT5 Polyclonal Antibody (Product # PA5-56100).

Target Information

Component of the DRB sensitivity-inducing factor complex (DSIF complex), which regulates mRNA processing and transcription elongation by RNA polymerase II. DSIF positively regulates mRNA capping by stimulating the mRNA guanylyltransferase activity of RNGTT/CAP1A. DSIF also acts cooperatively with the negative elongation factor complex (NELF complex) to enhance transcriptional pausing at sites proximal to the promoter. Transcriptional pausing may facilitate the assembly of an elongation competent RNA polymerase II complex. DSIF and NELF promote pausing by inhibition of the transcription elongation factor TFIIS/S-II. TFIIS/S-II binds to RNA polymerase II at transcription pause sites and stimulates the weak intrinsic nuclease activity of the enzyme. Cleavage of blocked transcripts by RNA polymerase II promotes the resumption of transcription from the new 3' terminus and may allow repeated attempts at transcription through natural pause sites. DSIF can also positively regulate transcriptional elongation and is required for the efficient activation of transcriptional elongation by the HIV-1 nuclear transcriptional activator, Tat. DSIF acts to suppress transcriptional pausing in transcripts derived from the HIV-1 LTR and blocks premature release of HIV-1 transcripts at terminator sequences. [UniProt]
