SRC3 Antibody (PA1-24638) in WB
Western blot analysis of SRC3 using nuclear extract (5µg for WB, 10 mg for CoIP) from HeLa cells. Samples were probed with a SRC3 polyclonal antibody (Product # PA1-24638) at a dilution of 0.33µg/ml for WB and 20µg/10 mg extract for CoIP. Detection was performed using Alkaline phosphatase conjugated goat anti-Rabbit IgG diluted 1:3,000 with NBT-BCIP as substrate for 20 min. WB and Coomassie Brilliant Blue R250 staining of SDS-PAGE gel followed by mass spectrometry to confirm the identity of the band that is SRC3 for CoIP.

The PA1-24638 immunogen is a synthetic peptide corresponding to a portion of human nuclear receptor coactivator 3 encoded within exon 16 (LocusLink ID 8202).
Target Information
AIB1-AC3 recognizes AIB1, a member of the SRC-1 family of nuclear receptor coactivators with an apparent molecular weight of 160kDa. The AIB1 gene has been mapped to region 20q12 and has been shown to be amplified in breast tumors and pancreatic cancer cell lines. AIB1 interacts in a ligand-dependent fashion with the estrogen receptor to enhance estrogen-dependent transcriptional gene activation.
