Sigma-1 Receptor Antibody (42-3300) in WB
Western blot of SIGMAR1 was performed by loading 30 µg of WT (lanes 1/3) and SIGMAR1 CRISPR KO (lanes 2/4) HEK293T or HeLa cell lysates in RIPA buffer onto a 5-16% gradient polyacrylamide gel. Proteins on the blots were visualized with Ponceau staining (below immunoblot). Proteins were transferred to nitrocellulose membrane and blocked in 5% milk for 1 hr. This antibody is specific for the sigma-1 receptor (sigma-1R, Oprs1 protein, opioid receptor sigma 1) protein. On western blots, it identifies the target band at ~25 kDa. Reactivity has been confirmed with rat brain, rat liver, and mouse brain homogenates. Based on amino acid sequence identity, reactivity with human, bovine, dog, and ermine is expected.
Target Information
This gene encodes a receptor protein that interacts with a variety of psychotomimetic drugs, including cocaine and amphetamines. The receptor is believed to play an important role in the cellular functions of various tissues associated with the endocrine, immune, and nervous systems. As indicated by its previous name, opioid receptor sigma 1 (OPRS1), the product of this gene was erroneously thought to function as an opioid receptor; it is now thought to be a non-opioid receptor. Alternative splicing of this gene results in transcript variants encoding distinct isoforms.
