Immunofluorescence analysis of Syndecan-2 was done on 90% confluent log phase Caco-2 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. The cells were labeled with Syndecan-2 Rabbit polyclonal Antibody (366200) at 2µg/mL in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor 488 Goat Anti-Rabbit IgG Secondary Antibody (A11008) at a dilution of 1:400 for 30 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant DAPI (S36938). F-actin (Panel c: red) was stained with Alexa Fluor 594 Phalloidin (A12381). Panel d is a merged image showing junctional localization. Panel e shows no primary antibody control. The images were captured at 20X magnification.

Syndecans are heparan sulfate proteoglycans (HSPGs) that are involved in cell adhesion, tissue morphogenesis, differentiation, and act as low affinity co-receptors of soluble growth factors. It is involved in the mitogenic activity and signaling of Granulocyte-Macrophage Colony-stimulating factor in osteoblasts, and has been shown to have a regulatory role in matrix assembly. Syndecan-2 is expressed at epithelial-mesenchymal interfaces, and prechondrogenic and preosteogenic mesenchymal condensations, which are areas of high morphogenetic activity.
