T7 RNA polymerase is a DNA-dependent RNA polymerase enzyme derived from the T7 bacteriophage. It is commonly used in molecular biology research and biotechnology applications. Expressed and purified from E. coli, T7 RNA polymerase functions by recognizing and binding to the T7 promoter sequence, initiating transcription of RNA from the corresponding DNA template. It has a single-subunit structure with distinct functional domains, including a DNA-binding domain, a catalytic domain, and a C-terminal domain. T7 RNA polymerase is highly processive, allowing for the synthesis of long RNA molecules without dissociating from the DNA template.
