TAP Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of TAP was performed using HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with Anti-TAP Polyclonal Antibody (Product # PA5-21415) at 1:50 dilution in 0.1% BSA and incubated overnight at 4 degree and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the composite image showing Nuclear with slight cytosolic localization of TAP in HeLa cells. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification with oil immersion.

TAP Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of paraffin-embedded NCIN87 Xenograft , using TAP (Product # PA5-21415) antibody at 1:100 dilution. Antigen Retrieval: EDTA based buffer, pH 8.0, 15 min.

TAP Antibody in Western Blot (WB) — Western blot was performed using Anti-TAP Polyclonal Antibody (Product # PA5-21415) and a 70 kDa band corresponding to TAP was observed across all cell lines tested. whole cell extracts (40 µg lysate) of HeLa (Lane 1), MCF-7 (Lane 2), A-431 (Lane 3), MOLT-4 (Lane 4), SH-SY5Y (Lane 5), SK-O-V3 (Lane 6), NIH/3T3 (Lane 7), Mouse Placenta (Lane 8), Mouse Heart (Lane 9), Mouse Kidney (Lane 10) and Rat Heart (Lane 11) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

TAP Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of the target protein. HeLa cells were transfected with TAP specific siRNA and decrease in signal intensity was observed in Immunofluorescence application using Anti-TAP Polyclonal Antibody (Product # PA5-21415). {KD}

Target Information

The constitutive transport element (CTE) of type D retroviruses serves as a signal of nuclear export for unspliced viral RNAs. TAP (also known as NXF1) mediates the export of CTE-containing simian type D retroviral RNAs through binding directly to the CTE. TAP is associated with a recognized mRNA export pathway and is a member of the multigene family of NXF proteins. NXF proteins belong to an evolutionarily conserved family of proteins, which are characterized by a leucine-rich-repeat domain (LRR) followed by a region known as the Nuclear Transport Factor 2 (NTF2)-like domain.
