TCF7 Antibody (PA5-77813) in WB
Western blot analysis of TCF7 in human cervical cancer cell line (HeLa) lysate using 10 µg of protein. Sample was prepared with 5% milk and TBST, incubated with TCF7 polyclonal antibody (Product # PA5-77813) using a dilution of 1:1000 (2 hours at RT) and followed by a Goat Anti-Rabbit HRP secondary antibody at a dilution of 1:3000 (1 hour at RT). Samples are arranged as follows: Lane 1: molecular weight ladder, Lane 2: Cervical Cancer cell line (HeLa) lysate.

A 1:1,000 dilution of PA5-77813 was sufficient for detection of TCF7 in 15 µg of human HeLa cell lysates by ECL immunoblot analysis using goat anti-rabbit IgG:HRP as the secondary antibody.|Detects 42 kDa, additional band at approximately 54 kDa is another isoform.
Target Information
TCF7 (transcriptional factor 7) is a transcriptional activator involved in T-cell lymphocyte differentiation. Necessary for the survival of CD4+ and CD8+ immature thymocytes. Isoforms for TCF7 lacking the N-terminal CTNNB1 binding domain cannot fulfill this role. TCF7 binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') found in the promoter of the CD3E gene. It may also act as feedback transcriptional repressor of CTNNB1 and TCF7L2 target genes. TLE1, TLE2, TLE3, and TLE4 repress transactivation mediated by TCF7 and CTNNB1.
