TDP1 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of paraffin-embedded human gastric, using TDP1 (Product # PA5-27111) antibody at 1:100 dilution. Antigen Retrieval: EDTA based buffer, pH 8.0, 15 min.

TDP1 Antibody in Western Blot (WB) — Western blot analysis of TDP1 was performed by loading whole cell lysate in 1X SDS sample buffer with 2-ME from 5 x 105 mouse embryonic fibroblasts (MEF). Samples were loaded onto a 4-12 % Bis-Tris polyacrylamide gel (Product # WG1402BOX). Proteins were transferred to nitrocellulose membrane with wet/tank transfer. Membrane was blocked in 5% milk/TBST. TDP1 was detected at approximately 60 kDa using a polyclonal anti-TDP1 antibody (Product # PA5-27111) at a dilution of 1:1000 in 5% milk/TBST at 4°C overnight, followed by a secondary antibody HRP-anti-rabbit at a dilution of 1:5000 at room temperature for 1 hour. Target Information

The protein encoded by this gene is involved in repairing stalled topoisomerase I-DNA complexes by catalyzing the hydrolysis of the phosphodiester bond between the tyrosine residue of topoisomerase I and the 3-prime phosphate of DNA. This protein may also remove glycolate from single-stranded DNA containing 3-prime phosphoglycolate, suggesting a role in repair of free-radical mediated DNA double-strand breaks. This gene is a member of the phospholipase D family and contains two PLD phosphodiesterase domains. Mutations in this gene are associated with the disease spinocerebellar ataxia with axonal neuropathy (SCAN1). While several transcript variants may exist for this gene, the full-length natures of only two have been described to date. These two represent the major variants of this gene and encode the same isoform.
