Immunofluorescent analysis of NCI-H460 cells using a TFAM polyclonal antibody (Product # PA5-23776). NCI-H460 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with a TFAM polyclonal antibody (Product # PA5-23776) (1:25, 1 hr at 37°C). Primary antibody was detected with fluor-conjugated donkey anti-rabbit secondary antibody (green) at 1:400 dilution for 50 min at 37°C). Actin filaments have been labeled with dye-conjugated phalloidin (red). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min).

This gene encodes a mitochondrial transcription factor that is a key activator of mitochondrial transcription as well as a participant in mitochondrial genome replication. Studies in mice have demonstrated that this gene product is required to regulate the mitochondrial genome copy number and is essential for embryonic development. A mouse model for Kearns-Sayre syndrome was produced when expression of this gene was eliminated by targeted disruption in heart and muscle cells.
