TOP3A Antibody (PA5-101160) in ICC/IF
Immunofluorescent analysis of TOP3A in HeLa cells. Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C), incubated with mouse anti-beta tubulin and TOP3A polyclonal antibody (Product # PA5-101160) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 (red) and goat anti-mouse IgG Alexa Fluor 488 (green).

Target Information
DNA topoisomerases are nuclear enzymes that regulate the topological structure of DNA by transiently breaking and rejoining DNA strands. Although DNA topoisomerase I (Topo I) and Topo II relax both positive and negative supercoils, Topo III relaxes only negative supercoils. Topo IIIalpha exists as a long and a short isoform, which are produced by alternative splicing. Topo IIIalpha, which localizes to the nucleolus, is constitutively expressed and remains at high levels throughout the cell cycle in HL-60 cells. Topo IIIbeta exists as three isoforms,beta-1,beta -2 and beta-3, also produced by alternative splicing. Topo IIIbeta-1 is expressed in testis, heart, and skeletal muscle, whereas Topo IIIbeta-2 is expressed in thymus, kidney and pancreas.
