TRADD Antibody (PA5-106869) in ICC/IF
Immunocytochemistry analysis of TRADD in A549 cells. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were incubated with TRADD polyclonal antibody (Product # PA5-106869) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies. The nuclear counter stain is DAPI (blue).

Antibody detects endogenous levels of total TRADD.
Target Information
TRADD (Tumor necrosis factor receptor type 1-associated DEATH domain protein) is an important mediator of FAS and TNFR1 associated apoptotic signaling. The nuclear form acts as a tumor suppressor by preventing ubiquitination and degradation of isoform p19ARF/ARF of CDKN2A by TRIP12. TRADD acts by interacting with TRIP12 and leading to the disruption of the interaction between TRIP12 and isoform p19ARF/ARF of CDKN2A. TRADD acts as an adapter molecule for TNFRSF1A/TNFR1 that specifically associates with the cytoplasmic domain of activated TNFRSF1A/TNFR1 mediating its interation with FADD. Overexpression of TRADD leads to two major TNF-induced responses - apoptosis and activation of NF-kappa-B.
