TTK Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry-Immunofluorescence analysis of of TTK in SH-SY5Y cells (green). Methanol fixed cells were blocked with 10% negative goat serum for 15 minutes at room temperature. Cells were probed with TTK Polyclonal Antibody (Product # PA5-119893) at a dilution of 1:50 for 1 hour at room temperature, washed with PBS. Alexa Fluor 488 conjugate-Goat anti-Rabbit IgG was used as the secondary antibody at 1:1,000 dilution. The nuclear counter stain is DAPI (blue).

TTK Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry (Paraffin) analysis of paraffin-embedded human breast carcinoma tissue using TTK Polyclonal Antibody (Product # PA5-119893). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the TTK antibody at a dilution of 1:100 for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

TTK Antibody in Western Blot (WB) — Western blot analysis of TTK on human placenta tissue lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. TTK Polyclonal Antibody (Product # PA5-119893) at 1:500 was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature. Predicted band size: 97 kDa Observed band size: 100 kDa.

TTK Antibody in Flow Cytometry (Flow) — Flow Cytometry analysis of TTK in MG-63 cells using TTK Polyclonal Antibody (Product # PA5-119893) at 1 µg/mL (red) compared with Rabbit IgG, monoclonal - Isotype Control (green). After incubation of the primary antibody at 4°C for 1 hour, the cells were stained with a Alexa Fluor 488 conjugate-Goat anti-Rabbit IgG Secondary antibody at 1:1,000 dilution for 30 minutes at 4°C (dark incubation). Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Target Information

This gene encodes a constant regulatory subunit of protein phosphatase 2. Protein phosphatase 2 is one of the four major Ser/Thr phosphatases, and it is implicated in the negative control of cell growth and division. It consists of a common heteromeric core enzyme, which is composed of a catalytic subunit and a constant regulatory subunit, that associates with a variety of regulatory subunits. The constant regulatory subunit A serves as a scaffolding molecule to coordinate the assembly of the catalytic subunit and a variable regulatory B subunit. This gene encodes an alpha isoform of the constant regulatory subunit A. Alternatively spliced transcript variants have been described.
