Immunofluorescent analysis of RFP Tag was performed using 70% confluent p65-mRFP transfected HEK-293 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. This antibody can be used to detect native RFP, RFP variants, and RFP fusion proteins in Western blot, immunoprecipitation, ELISA, and immunocytochemistry. This antibody has been shown to detect denatured and native TurboRFP, TurboFP602, TurboFP635, TagRFP, TagFP635, and mKate2.
Storage and reconstitution: to prepare a 1 mg/mL stock solution, add 100 µL sterile water or 50% glycerol to the vial. If desired, aliquot the antibody upon reconstitution to produce single-use samples. Store the reconstituted product with sterile water at 4° C, avoiding freeze/thaw cycles. When stored as directed, the solution is stable for 3 months. If reconstituted with 50% glycerol, store at -20° C, avoiding freeze/thaw cycles. When stored as directed, this solution is stable for 6 months.
Target Information
Red fluorescent protein (TagRFP) derived from Entacmaea quadricolor eqFP611 (Uniprot ID: Q8ISF8) is an important tool in biotechnology and cell biology as a spectrally distinct companion or substitute for the green fluorescent protein (GFP; from Aequorea jellyfish). Pigment protein. The emission does not change over the pH range of 4 to 10. Maturation occurs via a green intermediate and is complete within 12 hours. Recombinant expression at high temperatures induces oligomerization. Abs(max)=559 nm. Exhibits a smaller absorbance peak at 525 nm. Has a strong fluorescence emission spectrum which peaks at 611 nm.
