Heat-mediated antigen retrieval is recommended prior to staining, using an EDTA buffer for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is thymus tissue.
Terminal deoxynucleotidyl transferase (TdT) is a nuclear DNA polymerase responsible for the template-independent addition of N-nucleotides. It is expressed in normal hematopoietic cells, normal cortical thymocytes (immature, pre-B, pre-T lymphoid cells), and megakaryocytes of the bone marrow as well as acute lymphoblastic leukemia/lymphoma cells.
