Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is small intestine tissue.
Villin can cap, nucleate, sever and bundle actin in a Ca2+ and phosphoinositide-regulated manner. It is associated with the microvillar actin core bundle of intestinal and renal brush border. Villin is composed of six repeats, each containing 150 residues that together constitute the core domain. The core domain retains the Ca2+ dependent capping nucleating and severing activity, whereas the headpiece domain contributes towards actin filament bundling and binding F-actin.
